San is for people whom you are not familiar with. Chan is mostly for ladies but when you feel someone is adorable, you can use chan or its morphed version Tan. Kun is used primarily for males with exceptions as well. When someone calls you San, it is equivalent of sii (shi) which Koreans use to formally mention someone. They add nuna and hyung and other things when they get familiar.
